Method: The components were inserted into 24-well plates in a isolated or mixed forms and were immediately placed in contact with the culture medium (82.4 mm2 surface / ml) at 37 ° C with 100% humidity and 5% CO2) for 24h to prepare the original extract and the others 1:1 dilutions (1:2, 1:4, 1:8, 1:16, 1:32). The extracts were applied to culture of V79 fibroblast cells and cell viability was measured by MTT assay using spectrophotometer. The data were analyzed by the U-Mann-Whitney test (α = 0.05) (SPSS / PC +, version 11.5 SPSS, Chicago, IL, USA) and Dunn (5%).
Result: The RealSeal sealer showed to be most cytotoxic when compared with the sealer EndoRez, reducing cell survival in all dilutions. The EndoRez sealer promoted cell survival of 66.8%, 78.2% and 84.3% respectively in dilutions of 1:8, 1:16 and 1:32. For the separate components, it was observed that the component R of the RealSeal presented a slightly less cytotoxic than the W component, with up to 75% cell survival in the dilution 1:32. As for the separate components of EndoRez, the component W presented a cell survival of 71.7% in the 1:32 dilution, while the Y component maintained cell survival averaged 39% since the original extract 1:1 to the 1:32 dilution.
Conclusion: The sealer RealSeal both, with mixed or separate components, showed to be more cytotoxic than the EndoRez sealer in all forms.
